μ ªï Ë 17 Ë 1 - π 2553 1 π æπ åμâπ Original Article æ π ««Àå μ «Õ «Ÿ μâõß Õß««Àå å π Á â«õ μ ªÕ åøõ å π å «øï (Method Development and Validation for the Assay of Gliclazide Tablets by Ultra Performance Liquid Chromatography)»» Õ Ÿà ÿ * Abstract The analytical method for the analysis of Gliclazide in tablets was developed using Ultra performance liquid chromatography. The stationary phase was a ACQUITY UPLC TM BEH C18 (1.7 μm, 2.1 50 mm) at 40 degree Celsius, the molile phase was acetonitrile : the mixture of 0.1 ml of glacial acetic acid and 75 ml of water (27:73), the flow rate was 0.65 ml/min and detector wavelength was at 235 nm. The chromatogram showed Gliclazide peak at the retention time of about 4.8 minutes. The method validation data showed good specificity. The method was assessed by separation of Gliclazide from impurity F and its unknown degradation products under stress conditions; heat, light, acid, base and oxidation. The linearity test showed linearity relationship between the area under the peak and the concentration levels in range of 0.1-0.3 mg/ml with the correlation coefficient of 0.9997. The accuracy test showed that the percentage mean recovery of three concentration levels was 99.7 (%RSD = 0.80%). The method precision test or with in run showed good precision of the assay of six sample solutions (%RSD = 0.23). The intermediate precision of the assay of six sample solutions between days, analysts and instruments gave %RSD 0.67, 0.23 and 0.34, respectively. The method was rugged and robust, the variations in batch of column, column temperature, composition of mobile phase and flow rate did not affect the system suitability. Standard solution and sample solution were stable for 24 hours at room temperature in autosampler and for 96 hours in refrigerator. The related substance is one of the requirements for quality control of drug substances and drug products in the official pharmacopoeia. Nordiazepam is also an important metabolite of benzodiazepines excreted in urine. Nordiazepam Reference Standard is necessary for identification and assay of Nordiazepam in quality control of benzodiazepines products and determination of Nordiazepam content in urine samples and seized drug. Therefore, Bureau of Drug and Narcotic initiated the program for establishment of Nordiazepam Reference Standard. The candidate substance was synthesized. The identification test is performed by Infrared (IR) spectroscopy, Ultraviolet-visible (UV) spectrophotometry, Gas chromatography/mass spectrometry (GC/MS) and determination of the melting point. The results show that the samples are Nordiazepam. Moreover purity test utilized High performance liquid chromatography (HPLC) and Differencial scanning calorimeter (DSC) obtained purity value is 99.932 mol% and detection of any impurity and degradation product are not found. The assign value for quality test is evaluated by using non-aqueous acid base potentiometric titration and Clorazepate dipotassium monograph in the United States Pharmacopeia 29 (USP 29). Assign value and uncertainty result are 989.7 ± 2.6 μg/mg when can be traceable to the International System of Units (SI) by using potassium bipthalate NIST SRM batch 84k. Furthermore homogeneity testing are performed on randomized 20 bottles of samples both of between bottles and within bottles tests. From the results, it can be concluded that all samples are homogenized. Nordiazepam Reference Standard, packed in 50 mg per vial with label will be supplied with certificate of analysis for utilization in the Department of Medical Sciences, other reference laboratories and in private companies producing the benzodiazepines products. To be able to produce Nordiazepam Reference Standard helps to reduce the budget for purchasing the expensive reference standard from other countries, to improve the quality of products and to develop laboratory capability in both government and private sector. Garden Cress Seed is the dried mature seed of Lepidium sativum L.; Herbarium Specimen Number: DMSc 4533 (QSBG 22819); Crude Drug Number: DMSc 431 (Family Cruciferae). Constituents Garden Cress Seed contains sinapic acid, sinapine and imidazole alkaloids. It also contains volatile oil, fatty acids, amino acids, isothiocyanate glycosides, mucilage, etc. Description of the plant Annual herb, 20 to 80 cm high, erect, branched, nearly glabrous, glaucous. Basal leaves, pinnatisect or lyrate-pinnate, 2 to 10 cm long, 1 to 5 cm wide, segments toothed or finely divided, crispate, stalked to subsessile; stem-leaves becoming successively simple, linear and entire in the upper part. Inflorescence raceme, terminal or axillary; flowers small, 2 to 3 mm in diameter, bisexual, regular; sepals 4, oblong, 1 to 1.3 mm long, 0.5 to 0.8 mm wide; petals 4, white or lavender, spatulate or obovate, 2.5 to 4 mm long, 0.7 to 1.4 mm wide; stamens typically 6, tetradynamous; ovary superior, 2-carpeled, 1 ovule each, ripening into a thin partitioned pod.
